1354 Adult epidermal stem cells maintain their proliferative behavior in vitro

Whether epidermal stem cells (SCs) maintain their phenotype in vitro remains largely unknown. To determine whether adult human SCs self-renewal behavior vitro, we observed keratinocytes vitro. We studied live cell imaging of freshly obtained normal from surgical discards over 11-16 days. then tracked each colony and defined that divided as P (proliferating) did not divide (in 48h) D (differentiated). The three outcomes divisions were classified PP, PD, DD. able to define SCs, was done for neonatal (Roshan et al, 2016), by 100% PP the first 3 generations. formed significantly larger colonies, expected. Committed progenitor colonies mostly terminally differentiated (75%) during observation period, whereas no SC did. Over period observation, found dominated (82.8%±6.5), even at 10-14 generations, while committed only 30.6%±4.0 (p<0.0001).
